This. Konosuba has no illusions about the fact that its characters are terrible people. The comedy comes from their interactions. The plot is almost nonexistent, and is mainly an excuse for these four to still be in contact with each other.

Konosuba can basically be summed up as 'generic isekai protagonist gets sent to fantasy world to kill the demon king, refuses to leave the starting town because its safer there. Does low level adventures for drinking money by teaming up with a couple of other weirdos that no one else wants to deal with.'

Darkness herself is actually, weirdly, the most nuanced character of the bunch. Here you have this high-brow noble lady from a prestigious family who goes into the honorable profession of a lady crusader to hep the people of the realm. She actually takes her job really seriously when other people are in danger, and is perfectly willing to put herself in harms way to save others,
Unfortunately, it turns out that she is a HUGE masochist into pain and degradation. And when there isn't a situation that requires her to put on her serious face, she sort of lets her fantasies run wild to the point that more than one enemy has actually retreated from a fight because Darkness just made this way too weird for them.
One of the best sequences in the show involves her getting demonically possessed, and the demon quickly realizes that their whole plan is fucked sideways because Darkness is so singleminded in her weird obsessions that they keep losing control over her, she actually has more than enough willpower to resist the possession entirely. But they can't leave her and possess someone else because Darkness is so into the idea of being demonically possessed and forced to do stuff 'against her will' that she is actually keeping him from leaving.

Aka: Demon accidentally enters Darkness's magical realm and is unable to claw his way back out.

IIRC, Darkness's armor is actually some kind of adamantine-esq dense supermetal of the setting. Though Im pretty sure she just naturally has enough defense skills that she could tank most stuff naked.

I honestly think that the original plan was for Darkness to grow into the primary love interest role for Kazuma over time, a subversion having the last girl introduced be the romantic lead rather than the first one. She and Kazuma have a lot more meaningful personal interactions early on than the other characters, and at different times Kazuma admits that Darkness is almost his ideal girl and Darkness claims that a trashy person like Kazuma is just what she wants in a husband. And that whole fake marriage thing.

Of course, over time, Megumin gets moved into the main girl role as a result of character popularity.

I tried experimenting with a one-shot session where attack rolls were always determined by DEX, and damage rolls were always determined by STR. D&D 3.5 Crusader Class focused on the Devoted Spirit and White Raven disciplines.

Ask for DM permission to trade out one feat for a permanent effect of the Masochism spell from the Book of Vile Darkness (hah). This grants +1 to most d20 checks in the round after you take damage, scaling at +1 per 10 points of damage.

Crusader also gets a mechanic known as Steely Resolve, allowing them to delay the effect of a certain amount of damage in a round, granting a +1 bonus to attacks and damage in the round after they sink the damage. It starts out delaying 5 points and granting +1 at first level and caps out at 30 points of damage for +6 at Twentieth level.

Overall result, if Darkness hits 20th level and gets smacked with a Harm spell (150 Damage, fort-save for half) she could potentially get +6 from Steely Resolve and +12 from masochism to come out to +18 to fuckup whoever or whatever hit her, assuming she failed her saving throw for half. Alternatively, +6 from Steely Resolve and +7 from Masochism if she makes it.

In addition, the Devoted Spirit discipline of Martial Maneuvers (Weeaboo fightan techniques) is the paladin-ish one that lets you heal and protect your allies (which Darkness was devoted to in principle), while White Raven is usually about trying to charge in, leading the attack, and helping your allies to achieve victory rather than getting the killing blow yourself. You Can use Devoted Spirit to heal yourself as well, turning you into a damage-cycling constitution tank that sinks damage for your allies and draws enemy fire at every opportunity.

After that, nab the bodyguard feats that also help you jump into the path of danger for your allies.

The thief is supposed to be relatively competent, but often gets herself in trouble, and occasionally drags the party down with her. She doesn't play the role of a tank but a beater in combat. They are actually something of a mechanical genius, able to not just detect traps but determine how even the most complex trap functions. They can even build their own. They have more than enough skill to dismantle one.

In Vile Darkness there's a neat prestige class for fighters that expands the shit out of your crit range and gives some rust powers, as well as a shape shifting prestige class that I sort of like. Both of those I wouldn't use with the fluff as is since they require demon worshiping in a pretty childish sense.
As for exalted deeds... the parts I like aren't really mechanically sound. Vow of poverty is pretty useless or OP depending on your class (pretty much only good for druids). Some of the alternative animal companion/mount/familiar stuff might be good. I guess the non-lethal feats are okay, but I really dislike that they even have to be feats in the first place. There's an interesting template in there for "saints" or something like that which could potentially salvage a character with a shitty class. That's all I can think of off the top of my head as I haven't read any 3.5 books since 5e was a thing.
